The Effect of L-Theanine on Glycogen Phosphorylase b

Glycogen Phosphorylase b (GPb) is an enzyme that is imperative for energy metabolism. GPb alternates between its two forms, phosphorylase a (the active form) and phosphorylase b (the less-active form). This enzyme yields glucose-1-phosphate (G1P) and a glycogen polymer with one less glucose by cleaving a glucose molecule from the nonreducing ends of glycogen. GPb can have effectors that give it activating or inhibiting effects. In this study, the effect of L-Theanine was chosen as an effector to see what effect it has on the enzymatic activity of GPb, if any. L-Theanine is an amino acid that promotes relaxation, improves sleep, and has a range of health benefits. AMP, IMP, and epinephrine were investigated at low and high concentrations while assays were used to see the effect of the known activators IMP and epinephrine on GPb. To see the effects of L-Theanine being an activator or inhibitor, four assays were done. Results showed a slight inactivation of GPb because at a high concentration of effector, AMP showed a lower Km. One needs to understand the effect of L-Theanine on GPb as many take this supplement daily to help with their anxiety symptoms.
